Is the Sequenced Bordetella pertussis strain Tohama I representative of the species?
Subtractive hybridization was carried out to identify differences between the sequenced genome of Bordetella pertussis Tohama I and those of two recently collected isolates. We identified genetic regions specific to recent isolates, old isolates, and isolates of B. parapertussis and B. bronchiseptica species. We conclude that Tohama I strain is not representative of the B. pertussis species.
متن کاملDerivation of a physical map of the chromosome of Bordetella pertussis Tohama I.
We have used pulsed-field gel electrophoresis to derive a restriction map of the chromosome of Bordetella pertussis for the enzymes XbaI, SpeI, PacI, and PmeI, which cleave 25, 16, 2, and 1 times, respectively. The apparent size of the genome is 3,750 kb. The positions of genes for major virulence determinants in the vir regulon and of some housekeeping genes were determined. Apart from the pre...
متن کاملBordetella pertussis Isolates from Argentinean Whooping Cough Patients Display Enhanced Biofilm Formation Capacity Compared to Tohama I Reference Strain
Pertussis is a highly contagious disease mainly caused by Bordetella pertussis. Despite the massive use of vaccines, since the 1950s the disease has become re-emergent in 2000 with a shift in incidence from infants to adolescents and adults. Clearly, the efficacy of current cellular or acellular vaccines, formulated from bacteria grown in stirred bioreactors is limited, presenting a challenge f...
متن کاملBordetella pertussis Strain Lacking Pertactin and Pertussis Toxin.
A Bordetella pertussis strain lacking 2 acellular vaccine immunogens, pertussis toxin and pertactin, was isolated from an unvaccinated infant in New York State in 2013. Comparison with a French strain that was pertussis toxin-deficient, pertactin wild-type showed that the strains carry the same 28-kb deletion in similar genomes.
متن کاملExtraction of Outer membrane Vesicles from Vaccinal Strain of Bordetella Pertussis as the First Step of a Vaccine Candidate Study Against Pertussis Infection
Background: Pertussis is still one of the major public health problems. The increase of the disease emerged in recent decades due to the replacement of the reactogenic whole cell vaccine with the safer acellular vaccine and the genetic diversity of the bacterium. As outer membrane vesicles (OMVs) obtained from Bordetella pertussis contains surface immunogenic antigen in its structure, it has an...
